Description cDNA suppression subtraction library from oropharyngeal tonsils and mandibular lymph nodes of diseased and healthy European wild boar naturally exposed to M. bovis. Total RNA was isolated using TriReagent (Sigma, St. Louis, MO, USA) according to manufacturer's instructions. After diagnosis of diseased and healthy individuals by pathology, culture and spoligotyping of mycobacteria, pools of 20 ug RNA were made for healthy (N=2; 10 ug RNA/each) and diseased (N=4; 5 ug RNA/each) animals. SSH was performed at Evrogen JCS (Moscow, Russia) as previously described (Diachenko et al., 1996; 1999). Tester and driver RNAs were subtracted in both directions to construct 4 SSH libraries containing differentially expressed cDNAs in diseased (reverse-subtracted) and healthy (forward-subtracted) samples for oropharyngeal tonsils and mandibular lymph nodes. Approximately 100 clones from each library were randomly picked up and subjected to differential hybridization with subtracted and non-subtracted probes using the PCR-select differential screening kit (Clontech, Palo Alto, CA, USA), which resulted in >90% differentially expressed cDNAs.
